Overview

Adebrelimab + lenvatinib + chemotherapy is a combination therapeutic regimen being evaluated for the treatment of advanced biliary tract cancer (BTC). This regimen integrates three distinct therapeutic approaches: immune checkpoint inhibition, targeted anti-angiogenic therapy, and cytotoxic chemotherapy. Adebrelimab is a humanized monoclonal antibody that targets programmed death-ligand 1 (PD-L1), preventing its interaction with PD-1 to restore anti-tumor T-cell activity. Lenvatinib is an oral multi-kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR1-3), fibroblast growth factor receptors (FGFR1-4), and other tyrosine kinases, thereby inhibiting tumor angiogenesis and proliferation. The chemotherapy component, typically consisting of gemcitabine combined with cisplatin or oxaliplatin, induces direct tumor cell death and may promote an immunogenic tumor environment. This combination is primarily being investigated in clinical trials led by Harbin Medical University Cancer Hospital to improve outcomes in patients with unresectable or metastatic BTC.
