Overview
Adefovir dipivoxil is an orally administered acyclic nucleotide analog reverse transcriptase inhibitor (ntRTI) and prodrug of adefovir. It is used for the treatment of chronic hepatitis B virus (HBV) infection in patients aged 12 years and older. After oral administration, it is converted to its active form, adefovir diphosphate, by cellular kinases. The active metabolite inhibits HBV DNA polymerase (reverse transcriptase) by competing with deoxyadenosine triphosphate and causing DNA chain termination after incorporation into viral DNA. Adefovir dipivoxil does not cure hepatitis B but reduces viral replication and liver inflammation[1][3][4][5][6].
