Overview

Adegramotide (DSP-7888, also known as ombipepimut-S) is an investigational peptide-based cancer vaccine developed for the treatment of various hematologic malignancies and solid tumors, including glioblastoma multiforme and myelodysplastic syndromes. It is administered intradermally as a fixed-dose combination with nelatimotide. The vaccine contains synthetic peptides derived from the Wilms' tumor protein 1 (WT1), designed to stimulate both cytotoxic CD8+ T lymphocytes and helper CD4+ T cells against WT1-expressing tumor cells. Upon administration, antigen-presenting cells process these peptides for presentation on HLA molecules, activating WT1-specific T cells that migrate to the tumor microenvironment to exert antitumor effects. Clinical trials have shown immunogenicity in patients with higher-risk myelodysplastic syndrome who failed prior azacytidine treatment; however, several development programs have been discontinued due to insufficient efficacy in late-stage studies[1][3][4][5].
