adelbelimab + carboplatin + nab-paclitaxel

adelbelimab + carboplatin + nab-paclitaxel is an investigational chemo-immunotherapy regimen being evaluated as a neoadjuvant (preoperative) treatment for patients with resectable locally advanced head and neck squamous cell carcinoma (HNSCC). The regimen combines adelbelimab (KL-A167), a humanized IgG1 monoclonal antibody targeting programmed death-ligand 1 (PD-L1), with two chemotherapy agents: carboplatin (a platinum-based DNA alkylator) and nab-paclitaxel (an albumin-bound nanoparticle formulation of paclitaxel that stabilizes microtubules). By blocking the PD-1/PD-L1 axis, adelbelimab restores the anti-tumor activity of T-cells, while the chemotherapy components provide direct cytotoxic effects and potentially enhance the immunogenicity of the tumor. This combination is currently being studied in a Phase II trial sponsored by Sun Yat-Sen Memorial Hospital of Sun Yat-Sen University to assess its efficacy in achieving pathological complete response and improving surgical outcomes.
