Overview

Adeno-Slit2 is a replication-deficient adenoviral vector engineered to express the human Slit guidance ligand 2 (Slit2) protein. Developed primarily by researchers at The Ohio State University, Adeno-Slit2 is designed as an immunotherapeutic gene therapy for the treatment of advanced and metastatic breast cancer, as well as other solid tumors like gastric and colorectal cancers. Slit2 is a tumor suppressor that acts through the Roundabout homolog 1 (Robo1) receptor. In preclinical models, Adeno-Slit2 transduction overexpresses Slit2, which modulates the tumor microenvironment by inhibiting TGF-β1-mediated alternative activation of macrophages into pro-tumor tumor-associated macrophages (TAMs). This action suppresses downstream Smad2 and β-catenin signaling, thereby restricting tumor growth, promoting M1-like phagocytic macrophage activation, and reducing lung metastasis.
