adeno-TWIST1 vaccine

The adeno-TWIST1 vaccine (also known as Ad-Twist1 or Ad-Twist) is a recombinant adenovirus-based therapeutic cancer vaccine originally developed by Etubics Corporation (using their proprietary Ad5 [E1-, E2b-] platform) and subsequently developed by ImmunityBio and NantOmics. The vaccine utilizes a replication-defective adenovirus serotype 5 (Ad5) vector to deliver and express the basic helix-loop-helix transcription factor Twist1 (Twist), a well-characterized tumor-associated antigen and master regulator of epithelial-mesenchymal transition (EMT) that is overexpressed in various metastatic cancers. By expressing Twist1, the vaccine aims to stimulate a robust, antigen-specific CD4+ and CD8+ T-cell immune response to target and eliminate metastatic tumor cells. It has been evaluated in preclinical combination regimens with immunomodulators such as N-803 (an IL-15 superagonist), immune checkpoint inhibitors (e.g., anti-PD-L1, anti-CSF1R), and chemotherapies.
