Adenosine deaminase (ADA) is an enzyme involved in purine metabolism that catalyzes the deamination of adenosine to inosine and 2'-deoxyadenosine to 2'-deoxyinosine. It is used as enzyme replacement therapy for severe combined immunodeficiency disease (SCID) associated with adenosine deaminase deficiency. In ADA deficiency, adenosine and 2'-deoxyadenosine accumulate and are toxic to lymphocytes, leading to impaired immune function. ADA replacement therapy corrects these metabolic abnormalities, though improvement in immune function may take weeks to months after starting treatment.
