Adenosine diphosphate (ADP) is a naturally occurring purine nucleotide that plays a critical role in vascular biology and platelet function. In the context of research conducted at Colorado State University, ADP is utilized as an experimental pharmacological probe to investigate the signaling pathways of P2 purinergic receptors in humans. The study, titled Pyridoxine, P2 Receptor Antagonism, and ATP-mediated Vasodilation in Young Adults, examines how ADP and related nucleotides regulate blood flow, particularly during physiological stressors such as exercise and hypoxia. ADP acts as an agonist for several P2Y receptor subtypes, including P2Y1, P2Y12, and P2Y13, which are expressed on the vascular endothelium and smooth muscle cells. By administering ADP, researchers assess the efficacy of potential antagonists, such as Vitamin B6 (pyridoxine) or its metabolite pyridoxal-5-phosphate (PLP), in blocking these purinergic signaling pathways to better understand age-related declines in vascular function.
