Overview

Adenoviral p53 vector is a gene therapy product consisting of a replication-defective recombinant adenoviral vector carrying the wild-type human tumor suppressor protein p53 gene. It is designed to restore normal p53 function in cancer cells, leading to cell cycle arrest and apoptosis. The therapy works by introducing functional copies of the TP53 gene into tumor cells, which can induce growth arrest or programmed cell death in cells with dysfunctional or mutated endogenous TP53. Adenoviral p53 vectors have been evaluated as monotherapy and in combination with chemotherapy or radiation for various cancers, including non-small-cell lung cancer (NSCLC) and head and neck squamous cell carcinoma (HNSCC). Clinical studies have shown that these vectors are generally well tolerated, with fever being the most common side effect due to immune activation. Notable products include Gendicine (approved in China) and Advexin/INGN 201 (investigational). The mechanism involves direct tumor suppression via restored wild-type p53 activity as well as modulation of immune responses within the tumor microenvironment[1][2][3][4][5][6].
