Drug intelligence / Profile preview
Adenoviral vector expressing IFNγ and sTGFβRII is a dual-expressing recombinant adenoviral gene therapy developed by researchers at the University of Minnesota. It is designed to target pancreatic ductal adenocarcinoma (PDAC) by addressing both desmoplasia (fibrosis) and the highly immunosuppressive tumor microenvironment (TME). The vector delivers two therapeutic transgenes: interferon gamma (IFNγ), an immunostimulatory cytokine that promotes anti-tumor immune cell infiltration and activation, and the soluble extracellular domain of transforming growth factor-beta receptor II (sTGFβRII), which acts as a decoy receptor to block immunosuppressive and pro-fibrotic TGF-β signaling. In preclinical syngeneic mouse models of pancreatic cancer, intratumoral administration of this dual-expressing adenoviral vector demonstrated significant anti-tumor efficacy, reduced tumor fibrosis, increased immunogenicity, and elicited abscopal effects on untreated distant tumors.
