Adenovirus COVID-19 vaccines are a class of viral vector vaccines that utilize a modified, non-replicating adenovirus to deliver genetic material encoding the SARS-CoV-2 spike protein into human cells. Upon administration, the vector enters host cells and uses the cellular machinery to produce the spike protein, which then acts as an antigen to stimulate a robust immune response, including the production of neutralizing antibodies and the activation of T-cells. This platform has been employed by several developers globally, most notably CanSino Biologics (Ad5-nCoV), AstraZeneca/Oxford (ChAdOx1-S), Janssen/Johnson & Johnson (Ad26.COV2.S), and the Gamaleya Research Institute (Sputnik V). These vaccines have played a critical role in global vaccination efforts, particularly as heterologous booster doses following primary series vaccination with inactivated or mRNA vaccines.
