adenovirus-klk1 is an adenoviral-based gene therapy designed to deliver the human tissue kallikrein 1 (KLK1) gene. Developed by researchers at Temple University, it is being investigated for the treatment of systemic lupus erythematosus (SLE) and neuropsychiatric lupus (NPSLE). The mechanism involves the kallikrein-kinin system (KKS), where KLK1 expression leads to the suppression of Type I interferon (IFN) responses, reduction of plasma IFNα levels, and downregulation of interferon-responsive genes. In preclinical models, it has demonstrated the ability to ameliorate kidney pathology and improve depressive-like behaviors associated with lupus.
