Adenovirus-mediated human interleukin-12 is a gene therapy in which a recombinant adenoviral vector delivers the human interleukin-12 (IL-12) gene directly into tumor tissue or target cells. The expressed IL-12 protein acts as an immunomodulatory cytokine that stimulates the immune system by promoting activation of natural killer (NK) cells and cytotoxic T lymphocytes, and induces secretion of interferon-gamma (IFN-gamma). This results in enhanced antitumor immune responses and inhibition of tumor growth. The approach has been investigated primarily for cancer indications such as hepatic metastases from colon carcinoma and recurrent prostate cancer[1][3][4][5][6][7]. Preclinical studies have shown increased survival and tumor regression in animal models; early-phase clinical trials have demonstrated safety and immune activation.
