Overview

ETBX-071 (also known as Ad5-PSA or the adenovirus PSA vaccine) is a therapeutic cancer vaccine developed by ImmunityBio using a modified human adenovirus serotype 5 (hAd5) vector platform. The vaccine is engineered to express the prostate-specific antigen (PSA), a protein typically overexpressed in prostate cancer cells, to stimulate a targeted T-cell mediated immune response. The hAd5 platform used in ETBX-071 features deletions in the E1, E2b, and E3 regions, which reduces the expression of viral genes and minimizes the host's anti-vector immunity, potentially allowing for more effective repeated administrations even in patients with pre-existing adenovirus immunity. ETBX-071 is currently being evaluated in Phase 2 clinical trials for various stages of prostate cancer, including high-risk, recurrent, and hormone-refractory disease, often in combination with other immunotherapeutic agents such as N-803 (an IL-15 superagonist) and M-CENK (a yeast-based vaccine).
