Adenovirus serotype 5 cytomegalovirus murine interleukin-2 (Ad5-CMV-mIL-2) is a replication-deficient adenoviral gene therapy construct designed for the localized expression of murine interleukin-2 (mIL-2). The vector is based on the adenovirus serotype 5 (Ad5) platform, typically with E1 and/or E3 genomic deletions to prevent viral replication and increase payload capacity. Expression of the mIL-2 transgene is driven by the constitutive cytomegalovirus (CMV) immediate-early promoter, ensuring high-level production of the cytokine in transduced cells. In preclinical oncology research, this construct is primarily used to investigate the immunotherapeutic potential of IL-2 in mouse models of malignancy. Interleukin-2 acts as a potent growth factor for T cells and natural killer (NK) cells; by delivering the gene directly to the tumor microenvironment, researchers aim to stimulate a robust anti-tumor immune response while avoiding the severe systemic toxicities associated with high-dose recombinant IL-2 protein therapy.
