Adenovirus type 35 (Ad35) is a species B adenovirus being investigated as a viral vector for gene therapy and oncolytic applications. Unlike the more common species C adenoviruses (such as Ad5), Ad35 utilizes CD46 as its primary cellular receptor, which is ubiquitously expressed on human nucleated cells, including hematopoietic stem cells, dendritic cells, and natural killer (NK) cells. This unique tropism makes Ad35 a promising candidate for transducing immune cells for CAR-based therapies and for treating solid tumors where Ad5-based vectors may be limited by high pre-existing immunity or lack of the Coxsackie-adenovirus receptor (CAR). Research indicates that Ad35 exhibits potent oncolytic activity in adenocarcinoma cells and can efficiently deliver genetic payloads to NK cells.
