Overview

Adenovirus type 5 (Ad5) vaccine refers to a viral vector platform that utilizes a replication-deficient human adenovirus serotype 5 to deliver genetic material encoding specific antigens into host cells. The vector is typically engineered by deleting the E1 and/or E3 genomic regions, which renders the virus replication-incompetent and creates space for the insertion of foreign genes. Upon administration, the vector enters host cells—primarily via the coxsackievirus and adenovirus receptor (CAR)—and expresses the target antigen, thereby stimulating both humoral (antibody) and cellular (T-cell) immune responses. This platform has been employed for a wide range of pathogens, including HIV-1 (e.g., Merck's V520), Ebola virus (e.g., CanSino's Ad5-EBOV), and SARS-CoV-2 (e.g., CanSino's Convidecia). A major limitation of the Ad5 platform is the high level of pre-existing immunity to the vector in many human populations, which can neutralize the vaccine before it can induce a robust response to the encoded antigen.
