Adenylate kinase 1 (AK1) is a highly conserved enzyme that catalyzes the reversible phosphotransfer between two ADP molecules to generate ATP and AMP, thereby maintaining nucleotide homeostasis and supporting cellular energy dynamics. It is one of nine human isoenzymes and is extensively studied due to its implication in various diseases. Elevated AK1 activity has been associated with tau pathology and retinal inflammation, suggesting its inhibition could restore nucleotide homeostasis and alleviate these conditions. Deficiency in red blood cell AK1 is linked to congenital non-spherocytic hemolytic anemia and developmental delays. Its role is also being investigated in conditions like mucormycosis, cystic echinococcosis (as a vaccine candidate), right ventricular failure, male infertility, schistosomiasis, and acute myeloid leukemia.
