Aderbasib (INCB007839) is an orally bioavailable small molecule inhibitor of the ADAM (A Disintegrin And Metalloprotease) family, specifically targeting the sheddase activities of ADAM10 and ADAM17. By inhibiting these metalloproteases, aderbasib blocks the activation of epidermal growth factor receptor (EGFR) pathways and interferes with Notch signaling through cleavage inhibition. This mechanism may result in reduced tumor cell proliferation and has been investigated primarily as a targeted therapy for breast cancer, particularly HER2-positive metastatic disease. The drug was developed by Incyte and reached up to Phase 2 clinical trials before development was terminated[1][2][3][4][5][6][7][8].
