AdFITC-TCE is a preclinical, tandem single-chain variable fragment bispecific T-cell engager comprising a human anti-fluorescein E2 binding domain and a humanized anti-CD3 OKT3 binding domain. It binds CD3epsilon on T cells and fluorescein on tumor-bound antibody adaptors, thereby conditionally redirecting and activating T cells to lyse antigen-positive tumor cells. The approach has been evaluated with fluoresceinated CD33- and CD117-targeting adaptors in acute myeloid leukemia and with fluoresceinated rituximab and daratumumab adaptors in B-cell lymphoma and multiple myeloma models.
