Overview

ADG106 is a fully human, ligand-blocking, agonistic monoclonal IgG4 antibody that targets CD137 (also known as 4-1BB or TNFRSF9), a co-stimulatory receptor expressed on activated T cells and natural killer (NK) cells. By binding to a unique epitope of CD137, ADG106 acts as an agonist to stimulate immune cell activation and proliferation while also antagonizing the natural ligand of CD137. This dual mechanism enhances anti-tumor immune responses through increased cytokine secretion and cytolytic activity of T cells and NK cells. Preclinical studies have shown tumor suppression in various animal models, both as monotherapy and in combination with other immunotherapies such as PD-1/PD-L1 inhibitors. In clinical trials, ADG106 has demonstrated manageable safety up to 5 mg/kg IV infusion with preliminary efficacy in advanced solid tumors and non-Hodgkin's lymphoma[2][3][4][6][7].
