Overview

ADG116 is a fully human anti-CTLA-4 monoclonal antibody developed using Adagene’s NEObody technology. It targets a unique and highly conserved epitope of CTLA-4, enabling species cross-reactivity for robust preclinical studies. ADG116 acts through partial CD80/86 ligand blockade and enhanced regulatory T-cell (Treg) depletion via strong antibody-dependent cellular cytotoxicity (ADCC), resulting in immune activation with potentially improved safety and efficacy compared to existing anti-CTLA-4 therapies like ipilimumab. The drug is being evaluated as monotherapy and in combination with pembrolizumab for advanced/metastatic solid tumors, currently in phase 1b/2 clinical development.
