ADG138 is a novel bispecific T-cell engager (TCE) antibody designed to target both HER2 (ERBB2) and CD3. It integrates precision masking technology to control cytokine release syndrome and on-target off-tumor toxicity. The drug is intended for use as a single agent or in combination with other immune modulating agents for the treatment of HER2-expressing solid tumors. Preclinical data have shown potent anti-tumor activity in both high and low HER2-expressing tumors, including resistant/refractory tumors, with an excellent safety profile—demonstrating a 100-fold reduction in cytokine release compared to its parental non-masked TCE[1][4][6].
