ADG153

ADG153 is a fully human anti-CD47 monoclonal antibody prodrug developed using Adagene's proprietary SAFEbody precision masking technology. This technology conditionally masks the antibody with activatable peptides, allowing it to remain inactive in normal tissues and become activated specifically within the protease-rich tumor microenvironment. Upon activation, ADG153 binds to CD47 on tumor cells, blocking the CD47/SIRPα signaling pathway and promoting immune-mediated tumor cell destruction. The IgG1 format of ADG153 demonstrates potent anti-tumor activity through both antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP), while minimizing red blood cell-related toxicities and antigen sink effects that are common with other anti-CD47 therapies. Preclinical studies have shown strong efficacy in solid tumor models, extended half-life compared to benchmark antibodies, and a favorable safety profile.
