Overview

ADG206 is a masked, IgG1 Fc-engineered anti-CD137 (4-1BB) monoclonal antibody developed by Adagene for cancer immunotherapy. It utilizes precision masking technology to conditionally activate the antibody in the tumor microenvironment, thereby limiting on-target off-tumor toxicities. Upon activation in tumors, it binds strongly to CD137 (a co-stimulatory receptor on activated T cells), promoting T cell activation and antitumor immune responses through heightened FcγR-mediated crosslinking. Preclinical studies have shown robust single-agent antitumor activity and enhanced efficacy when combined with other checkpoint inhibitors such as anti-PD-1 or anti-CTLA-4 therapies. The drug is currently being evaluated in a phase 1 clinical trial for patients with advanced or metastatic solid tumors[1][3][6].
