AdGFPFasL is a replication-incompetent adenoviral vector designed for gene therapy applications, primarily investigated in oncology. This construct delivers two genes: Green Fluorescent Protein (GFP), which serves as a marker for tracking gene expression and viral transduction, and a modified murine Fas Ligand (FasL). The therapeutic mechanism of AdGFPFasL relies on the expression of FasL within target cells. FasL is a protein that binds to the Fas receptor (FasR) on the surface of cells, initiating the extrinsic apoptotic pathway and leading to programmed cell death. This action has been shown to induce apoptosis in cancer cells, including those resistant to anti-Fas monoclonal antibodies, and can also elicit a 'bystander effect' where uninfected neighboring tumor cells are also eliminated. Research has explored its potential in treating head and neck squamous cell carcinoma and prostate cancer.
