ADGN-901 is a preclinical-stage peptide-mRNA nanoparticle therapeutic designed to restore wild-type BRCA1 tumor suppressor function in BRCA1-deficient or mutated cancers, such as triple-negative breast cancer. Developed by Aanastra (formerly Aadigen) in collaboration with DivinCell, the therapeutic consists of a proprietary BRCA1-derived mRNA complexed with short, tumor-selective amphipathic peptides. This peptide-based nancarrier platform facilitates systemic, non-endocytic intracellular delivery of the mRNA, leading to the rescue of BRCA1 expression and restoration of homologous recombination DNA repair. In preclinical models, ADGN-901 has demonstrated significant tumor growth inhibition and has been shown to resensitize PARP inhibitor-resistant cancer cells to therapies like veliparib.
