ADGN-903 is a preclinical-stage mRNA-peptide nanoparticle therapeutic designed to restore wild-type BRCA1 tumor suppressor function in BRCA1-mutated or deficient cancers. Developed by Aanastra (formerly Aadigen) in collaboration with Divincell, the drug utilizes a proprietary delivery platform consisting of short amphipathic peptides that complex with BRCA1-derived mRNA to form stable, tumor-selective nanoparticles. By delivering functional BRCA1 mRNA, ADGN-903 rescues homologous recombination DNA repair pathways, inhibits tumor cell proliferation, and prevents RAD51 recruitment to DNA damage sites. In preclinical models of triple-negative breast cancer, ADGN-903 has demonstrated strong anti-tumor efficacy and the ability to resensitize PARP inhibitor-resistant tumors to therapies like veliparib.
