ADGP75 is an adenoviral vector-based investigational therapy designed to express the murine melanoma antigen gp75. It is used in preclinical studies to induce an immune response against melanoma cells, primarily in mouse models. The mechanism involves gene transfer of gp75 antigen via an adenovirus (Ad) vector, leading to immune activation and tumor rejection. Mechanistically, ADGP75 immunization stimulates cellular immune responses (CD4+ and CD8+ T-cell mediated) rather than a humoral response, enabling rejection of melanoma cells. It has primarily been studied in C57BL/6 mice with established B16 melanoma pulmonary metastases and is considered an immunotherapeutic strategy targeting the melanocyte differentiation antigen gp75 for metastatic melanoma[1][3][5][7].
