ADGRE2-CAR-28z1XX is a chimeric antigen receptor (CAR) component designed to target the ADGRE2 protein (also known as EMR2), an adhesion G protein-coupled receptor expressed on myeloid cells and frequently overexpressed in acute myeloid leukemia (AML). Developed by researchers at Memorial Sloan Kettering Cancer Center, this CAR construct features a second-generation design with a CD28 costimulatory domain and a modified CD3ζ signaling domain. The modification, termed "1XX", involves mutating two of the three immunoreceptor tyrosine-based activation motifs (ITAMs) in the CD3ζ chain to be non-functional, leaving only the membrane-proximal ITAM active. This design is intended to calibrate T-cell activation, reducing exhaustion and enhancing the in vivo persistence and functional fitness of the CAR-T cells. ADGRE2-CAR-28z1XX is typically employed as the primary signaling unit within the combinatorial CAR platform ADCLEC.syn1, which pairs it with a CLEC12A-targeting chimeric costimulatory receptor (CCR) to achieve logic-gated (IF-BETTER) targeting of AML cells while minimizing toxicity to normal hematopoietic stem and progenitor cells.
