ADGRE2 CAR-T is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by Zhejiang University for the treatment of relapsed or refractory acute myeloid leukemia (AML). The therapy involves the genetic modification of a patient's own T cells to express a CAR that targets ADGRE2 (Adhesion G Protein-Coupled Receptor E2, also known as EMR2 or CD312). ADGRE2 is a member of the adhesion G protein-coupled receptor family that is highly expressed on AML blasts and leukemic stem cells, while showing minimal expression on normal hematopoietic stem cells, making it a suitable target for reducing off-tumor toxicity. The therapy is currently being evaluated in a Phase I clinical trial (NCT05748197). Advanced research iterations include a sensitivity-tuned or "IF-BETTER" gated CAR-T (ADCLEC.syn1) which incorporates a CLEC12A-targeted chimeric costimulatory receptor to further refine specificity against leukemic stem cells.
