ADH-1 + carboplatin is an investigational combination therapy consisting of ADH-1 (Exherin), a small molecule competitive inhibitor of N-cadherin, and carboplatin, a platinum-based alkylating chemotherapeutic agent. ADH-1 targets N-cadherin on tumor cells and vasculature to disrupt cell adhesion and induce tumor vascular angiolysis and apoptosis. It has shown potential anti-tumor activity in preclinical models and early clinical studies for tumors expressing N-cadherin. Carboplatin exerts its cytotoxic effect by forming DNA crosslinks that inhibit DNA replication and transcription, leading to cell death. The combination is being evaluated in early-phase clinical trials for synergistic anti-tumor effects in various solid tumors[1][2][4].
