AdhAQP1 is a first-generation, E1-deleted, replication-defective serotype 5 adenoviral vector that encodes the cDNA for the water channel protein human aquaporin‑1 (hAQP1). It is designed for gene therapy by transfecting salivary gland cells to express hAQP1 in their apical and basolateral membranes. The primary indication has been the treatment of radiation-induced salivary hypofunction (xerostomia) in patients with head and neck cancer. By restoring water permeability to damaged acinar cells, it aims to increase fluid secretion from irradiated salivary glands. Clinical trials have shown increased parotid flow and improved xerostomic symptoms in some patients after a single administration, with effects lasting several weeks to years in responders. The drug was initially developed by the National Institute of Dental & Craniofacial Research (NIDCR), part of NIH[3][4][6][7][9].
