ADI-002

ADI-002 is an investigational allogeneic Vδ1 gamma delta CAR-T cell therapy engineered to target glypican-3 (GPC3) and armored with a cell-intrinsic soluble interleukin-15 (IL-15) to enhance expansion, persistence, and antitumor activity in solid tumors. Preclinical data showed potent, specific cytotoxicity against GPC3-positive cancer cell lines, robust cytokine release, and strong in vivo antitumor activity in hepatocellular carcinoma models without observed graft-versus-host effects. The product demonstrates high expression of innate activating receptors (NKG2D, DNAM-1) with low expression of exhaustion markers (PD-1, LAG-3, TIGIT, TIM-3). ADI-002 is being developed primarily for GPC3-positive solid tumors such as hepatocellular carcinoma by Adicet Bio, with IND-enabling work reported and preclinical development status noted. Manufacturing uses a gamma-retroviral vector encoding the GPC3 CAR and secreted IL-15, aligned with Adicet’s established γδ T-cell platform.[1][4][7][3]
