ADI-270 is an investigational, allogeneic, off-the-shelf gamma delta (γδ) chimeric antigen receptor (CAR) T cell therapy designed to target CD70-positive cancers. It utilizes Vδ1 γδ T cells engineered with a third-generation CAR that employs the natural CD27 receptor as the binding domain for CD70. The therapy is further "armored" by expressing a dominant negative form of transforming growth factor-beta receptor II (dnTGFβRII), which confers resistance to immunosuppressive signals in the tumor microenvironment. This design aims to enhance anti-tumor activity, persistence, and reduce susceptibility to host immune rejection. ADI-270 has demonstrated potent preclinical activity against both solid and hematological malignancies expressing CD70, including clear cell renal cell carcinoma (ccRCC), non-small cell lung cancer, and T-cell lymphoma. It is currently being evaluated in phase 1/2 clinical trials for relapsed or refractory ccRCC after prior treatment with immune checkpoint inhibitors and VEGF inhibitors[1][2][3][4][5][8].
