ADI-925 is an allogeneic off-the-shelf gamma delta T cell therapy engineered with a Chimeric Adapter (CAd) to enhance recognition of tumor stress ligands and amplify innate and adaptive antitumor signaling. It leverages native receptors such as NKG2D and stress ligand pathways (MICA, MICB, ULBP1-6) via a modified DAP10 signaling adapter to boost cytotoxicity and persistence without adding an extracellular CAR domain. Preclinical data show robust cytotoxic and proliferative activity, sustained NKG2D expression, favorable cytokine profiles, and in vivo tumor control with kinetics similar to CAR T cells, supporting potential application across solid tumors and hematologic malignancies. Developed by Adicet Bio; an IND had been planned for H2 2023, and external reporting indicates the company later paused this preclinical program while prioritizing other assets.[2][6][5][1][7][3]
