ADI-PEG 20 + docetaxel

ADI-PEG 20 + docetaxel is a combination therapy consisting of pegylated arginine deiminase (ADI-PEG 20) and the chemotherapeutic agent docetaxel. ADI-PEG 20 is an enzyme that depletes extracellular arginine by converting it to citrulline, exploiting the metabolic vulnerability of cancer cells that lack argininosuccinate synthetase (ASS1) and are therefore unable to synthesize arginine. This leads to selective starvation and death of ASS1-deficient tumor cells. Docetaxel is a microtubule inhibitor that disrupts cell division by stabilizing GDP-bound tubulin in the cell, thereby inhibiting mitosis. The combination has shown synergistic antitumor activity in preclinical models and early-phase clinical trials for advanced solid malignancies, including castrate-resistant prostate cancer and non-small cell lung cancer[2][4][5]. Expansion cohorts have been conducted or are ongoing for these indications at recommended phase II doses.
