**Adibelivir (IM-250)** is an orally active small molecule helicase-primase inhibitor developed by Innovative Molecules for treating herpes simplex virus (HSV) infections. It potently inhibits HSV-1 replication (IC50 20 nM) by targeting the viral helicase-primase complex, blocking DNA synthesis, and demonstrates superior nervous system penetration (brain/plasma ratio 0.5-4) compared to other inhibitors like pritelivir and amenamevir. Preclinical data show efficacy in reducing disease duration, preventing recurrences, limiting viral shedding, and treating herpes encephalitis and neonatal models by impairing latent reservoir reactivation.[1][3][5][10][11]
