Drug intelligence / Profile preview
Adilipoline (CL413) is a water-soluble, chimeric, dual-acting toll-like receptor 2 (TLR2) and toll-like receptor 7 (TLR7) agonist developed by InvivoGen. It is chemically composed of the TLR7 ligand 8-hydroxyadenine conjugated to the terminal acid function of the TLR2 ligand Pam2CSK4. By co-activating both cell-surface TLR2 and endosomal TLR7, adilipoline triggers synergistic innate immune signaling pathways, inducing both NF-κB and IRF activation. This dual activation leads to robust production of pro-inflammatory cytokines and type I interferons. Adilipoline is primarily utilized as a vaccine adjuvant to enhance both humoral and cellular immune responses, showing promise in preclinical models of influenza A, respiratory syncytial virus (RSV), and Chikungunya virus infections. Additionally, it has demonstrated potent antitumor activity, promoting tumor regression upon intratumoral administration in melanoma models.
