Overview
Adipiplon is a nonbenzodiazepine anxiolytic and investigational small molecule drug developed by Neurogen Corporation. It acts as a highly selective partial agonist at the Gamma-aminobutyric acid receptor subunit alpha 3 (GABRA3), with strong preference for the α3 subtype and minimal affinity for α1, α2, or α5 subtypes. This selectivity distinguishes it from other nonbenzodiazepine anxiolytics and was intended to provide anxiolytic and sedative effects with fewer side effects than traditional benzodiazepines. Adipiplon was investigated primarily for insomnia, anxiety disorders, and schizophrenia. Clinical trials reached Phase II before development was discontinued due to significant next-day side effects[1][3][4][7].
