Overview

An adjuvanted high dose quadrivalent VLP (virus-like particle) vaccine is an investigational or developmental influenza vaccine designed to provide enhanced protection against four strains of influenza virus (two A and two B lineages). The "high dose" refers to a higher amount of hemagglutinin antigen per strain compared to standard-dose vaccines, aiming to elicit a stronger immune response, particularly in populations with weaker immunity such as older adults. The "adjuvanted" component means the formulation includes an immunostimulatory substance (commonly aluminum salts or other novel adjuvants) that further boosts the immune response by recruiting antigen-presenting cells and enhancing both humoral and cellular immunity. The VLP modality mimics the structure of native viruses without containing viral genetic material, making it non-infectious but highly immunogenic. Plant-derived versions have been developed using transient expression systems in Nicotiana benthamiana. Clinical studies have shown that these vaccines can induce robust antibody and T-cell responses at both low and high doses, with alum-adjuvant formulations further increasing immunogenicity[3][5]. These vaccines are being developed primarily for seasonal influenza prevention in adults—including those over 50 years old—where traditional vaccines may be less effective.
