Overview

The adjuvanted quadrivalent cell-based inactivated influenza vaccine (high-dose), also known as aHD-QIVc, is an experimental vaccine candidate developed by Seqirus (a CSL company) for the prevention of seasonal influenza in adults aged 65 and older. This formulation is unique in that it combines three distinct enhancement technologies: a quadrivalent antigen profile, a cell-based manufacturing process using MDCK cells (which avoids egg-adaptive mutations), and a high-dose antigen concentration (60 µg of hemagglutinin per strain) combined with the MF59 adjuvant (a squalene-based oil-in-water emulsion). By combining increased antigen content with an adjuvant, the vaccine aims to elicit a more robust and broader immune response in the elderly, a population often characterized by immunosenescence and reduced vaccine efficacy. It was evaluated in Phase 2 clinical trials (e.g., NCT04326062) to compare its safety and immunogenicity against other enhanced influenza vaccine platforms, such as standard-dose adjuvanted and high-dose non-adjuvanted vaccines.
