ADL 10-0101

ADL 10-0101 is a small molecule, peripherally selective kappa opioid receptor agonist developed as an analgesic. It does not cross the blood-brain barrier at therapeutic doses, which is expected to minimize central nervous system side effects commonly associated with opioid therapy. Preclinical and early clinical studies suggested efficacy in treating inflammatory pain, visceral pain (such as that seen in chronic pancreatitis), burn pain, and dermal itch. The drug was investigated for both intravenous infusion and potential topical administration. Clinical trials included Phase I safety studies and Phase II proof-of-concept trials for visceral pain and itch; however, development was ultimately discontinued[1][2][3][4][5][7][9].
