ADL7445 is a proprietary, small molecule, peripherally-acting mu-opioid receptor antagonist (PAMORA) that was developed for the treatment of chronic opioid-induced constipation (OIC). Originally discovered by Eli Lilly and Company, the compound was acquired by Adolor Corporation in 2009. ADL7445 is designed to selectively antagonize mu-opioid receptors in the periphery, particularly the gastrointestinal tract, to mitigate the side effects of opioid therapy such as bowel dysfunction. Because it does not readily cross the blood-brain barrier, it aims to provide relief from constipation without reversing the central analgesic effects of opioids. The drug entered Phase I clinical development in 2009, but its development was discontinued following the acquisition of Adolor by Cubist Pharmaceuticals.
