AdML-VTIN is a transductionally-targeted oncolytic adenovirus (Ad) designed for the treatment of mesothelin (MSLN)-expressing cancers, particularly pancreatic cancer. Developed by researchers at the University of Minnesota, the virus features a redesigned fiber AB-loop that specifically targets mesothelin, a protein highly overexpressed in pancreatic adenocarcinoma. AdML-VTIN was isolated from a high-diversity adenovirus library through high-throughput screening for infectivity in MSLN-expressing cells. In preclinical models, the virus demonstrated significant antitumor activity and selective replication in MSLN-positive tumors. Notably, AdML-VTIN exhibits reduced liver sequestration and enhanced tumor delivery following systemic administration compared to wild-type Ad5, potentially overcoming major hurdles for the systemic delivery of oncolytic gene therapies.
