Adn-IGF1R is a targeted biologic belonging to the Adnectin class, which are proteins derived from the human fibronectin tenth type III domain (10Fn3). Developed by Adnexus (a Bristol-Myers Squibb company), Adn-IGF1R is engineered to bind with high affinity to the extracellular domain of the insulin-like growth factor 1 receptor (IGF1R). The insulin-like growth factor signaling pathway is a key regulator of cellular proliferation and survival, and IGF1R is frequently overexpressed in various cancers, including breast, pancreatic, and ovarian tumors. Adn-IGF1R acts by interfering with IGF1R signaling, thereby inhibiting downstream pathways such as AKT phosphorylation and reducing tumor cell growth. In preclinical research, it has also been evaluated as a targeting component in fusion proteins (e.g., with recombinant gelonin) to deliver cytotoxic payloads specifically to IGF1R-expressing cells.
