Overview

AdNRGM is an investigational gene therapy consisting of a recombinant adenoviral vector (typically serotype 5) encoding the epidermal growth factor (EGF)-like domain of human neuregulin-1 (NRG1). Developed by Zensun (Shanghai) Sci & Tech, it is designed for the treatment of chronic heart failure. The mechanism involves the local expression of NRG1, which binds to and activates ErbB4 receptors on cardiomyocytes, subsequently inducing heterodimerization with ErbB2. This activation triggers downstream signaling pathways, including PI3K/Akt and MAPK, which promote cardiomyocyte survival, improve sarcomere organization, and enhance cardiac contractility. By delivering the NRG1 gene directly to the cardiac environment, AdNRGM aims to provide a sustained therapeutic effect and overcome the short half-life limitations associated with recombinant protein administration.
