Overview

ADO09 is a ready-to-use, fixed-ratio co-formulation of pramlintide (the only FDA-approved amylin analogue) and the insulin analogue A21G (also known as M1, the main circulating metabolite of insulin glargine). Developed for subcutaneous injection before meals, it is intended to improve postprandial glucose control and long-term outcomes in people with type 1 diabetes by combining two complementary hormones—insulin and amylin analogues—in a single formulation. Clinical studies have shown that ADO09 significantly reduces postprandial blood glucose excursions compared to insulin lispro or aspart alone, improves time-in-range for glycemic control without increasing hypoglycemia risk, slows gastric emptying similar to separate pramlintide administration, and may contribute to weight reduction. The drug has been well tolerated in clinical trials with transient gastrointestinal side effects being the most common adverse events[1][6][8][9].
