ADoBind-DM1 is a first-in-class tetravalent antibody-drug conjugate (ADC) targeting folate receptor alpha (FRα). Developed by Medicovestor and Sanyou Biopharmaceuticals, it utilizes the ADoBind platform, which consists of a tetravalent antibody backbone created by crosslinking two monomeric IgG1 molecules via a disulfide bond (S444C mutation). This specific ADC features a noncleavable SMCC linker conjugated to the microtubule inhibitor DM1 (mertansine) with an average drug-to-antibody ratio (DAR) of 8. ADoBind-DM1 is designed to treat FRα-expressing malignancies, such as pancreatic ductal adenocarcinoma (PDAC) and ovarian cancer, by combining potent payload-mediated cytotoxicity with enhanced antibody-dependent cellular cytotoxicity (ADCC) due to its tetravalent structure. In preclinical studies, it demonstrated superior avidity, pharmacokinetic properties, and tumor growth inhibition compared to monomeric counterparts.
