Overview

Adomeglivant (LY2409021) is a potent and selective small-molecule antagonist of the glucagon receptor. Developed by Eli Lilly, it was investigated as an oral treatment for type 2 diabetes mellitus. By blocking the glucagon receptor, adomeglivant reduces hepatic glucose production, leading to lower blood glucose levels in both healthy individuals and those with type 2 diabetes. Clinical studies demonstrated significant reductions in HbA1c and fasting plasma glucose with good overall tolerability and a low risk of hypoglycemia. However, treatment was associated with reversible increases in serum aminotransferases and hepatic fat content. Development was discontinued after phase 2 clinical trials[1][4][5][6][7].
